Application Notes
Applications Validated by Active Motif:
IP: 10 µl per IP
WB: 1:500 - 1:1,000 dilution
Immunogen
This FOXO1/FKHR antibody was raised against a peptide from the C-terminal region of human FOXO1/FKHR.
Buffer
Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.
FOXO1/FKHR antibody (pAb) tested by Immunoprecipitation.
10 µl of FOXO1/FKHR antibody was used to immunoprecipitate FOXO1/FKHR from 500 µg of K562 nuclear cell extract (lane 2). 10 µl of rabbit IgG was used as a negative control (lane 1). The immunoprecipitated protein was detected by Western blotting using the FOXO1/FKHR antibody at a dilution of 1:500.
FOXO1/FKHR antibody (pAb) tested by Western blot. U2OS nuclear extract (20 µg per lane) was probed with 39629 at a 1:500 dilution.
Background
FOXO1/FKHR (forkhead box 01) belongs to the forkhead family of transcription factors that contain a distinct forkhead domain. Chromosomal aberrations involving FOXO1, such as translocation with PAX3, are associated with aveolar rhabdomyosarcoma. The specific function of this gene is undetermined.
Positive Control
Nuclear extracts from U2OS cells can be used as a positive control for FOXO1/FKHR antibody.
