Sample preparation is a critical step for any assay, and making sure nuclei are intact for CUT&RUN or CUT&Tag assays is key for successful results. The Tissue Prep for NGS Assays kit is optimized for processing tissue to yield nuclei for ChIC/CUT&RUN and CUT&Tag assays.

Tissue Prep for NGS Assays includes Lysis Buffer, Protease Inhibitor Cocktail, Strainers, and 1 X Wash Buffer optimized to yield intact nuclei for the ChIC/CUT&RUN and CUT&Tag-IT® R-loop Assays for the preparation of 24 samples. These components are also in the original CUT&Tag-IT® Assay Kit – Tissue (16 rxn).

10 mg mouse liver tissue were processed with Tissue Prep for NGS Assays. Isolated nuclei were obtained and 500,000 nuclei were used in duplicate CUT&RUN reactions targeting either YY1 or JunD. Libraries were generated with DNA Library Prep Kit for Illumina® Cat. No. 53220 and Dual Index Primers Set 1 Cat. No. 53221.
