Applications

Application Notes
Applications Validated by Active Motif:
WB: 1:500 dilution
Ikaros may be present as multiple protein isoforms due to alternative splicing. Commonly, the two largest isoforms are detected as seen in the example Western blot image.
Published Applications
The following applications have been published using this antibody. Unless noted above, Active Motif may not have validated the antibody for use in these applications:
ChIP, ChIP-Seq
EMSA

Immunogen
This Ikaros antibody was raised against a recombinant protein corresponding to the N-terminal half of mouse Ikaros.
Buffer
Rabbit serum containing 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.
Ikaros pAb tested by Western blot.
Detection of Ikaros by Western blot analysis. Ikaros was detected using a 1:1,000 dilution of Ikaros pAb with 20 µg mouse spleen tissue extract.
Background
Ikaros is a transcription factor that plays a critical role in the control of lymphohematopoiesis and immune regulation. All Ikaros isoforms share a common C-terminal domain that contains a transcription activation motif and two zinc finger motifs required for hetero- and homo-dimerization among the Ikaros isoforms, and for interactions with other proteins. Ikaros is abundantly expressed in pituitary mammosomatotrophs, where it deacetylates Histone H3 sites on the proximal growth hormone (GH) promoter to silence gene expression.
Positive Control
Whole-cell extracts from mouse spleen can be used as a positive control for Ikaros antibody.
Chromatin IP Assays
This antibody has been validated for use in ChIP and/or ChIP-Seq, and can be used with Active Motif's ChIP-IT® High Sensitivity Kit or our magnetic bead-based ChIP-IT® Express Kits. For an overview of all of our ChIP products, please visit our Chromatin IP Page to learn about ChIP products designed for use with Ikaros antibody (pAb).
Storage
Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20°C for up to 2 years. Keep all reagents on ice when not in storage.
