Background
UBE2L3 (Ubiquitin-conjugating enzyme E2 L3) also known as E2-F1, L-UBC, UBCH7 and UbcM4. UBE2L3 that specifically acts with HECT-type and RBR family E3 ubiquitin-protein ligases. It has activity with the RBR family E3 enzymes, such as PRKN and ARIH1, that function like RING-HECT hybrids. Accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. In vitro catalyzes ‘Lys-11’-linked polyubiquitination. Involved in the selective degradation of short-lived and abnormal proteins. Down-regulated during the S-phase it is involved in progression through the cell cycle. Regulates nuclear hormone receptors transcriptional activity. May play a role in myelopoiesis.

Protein Details
Recombinant UBE2L3 protein that includes full length of human UBE2L3 protein (accession number NP_003338.1) was expressed in E. coli and contains an N-terminal His tag with a molecular weight of 20.02 kDa. The purity of the protein is ≥ 95% by SDS-PAGE.

AMP-Glo assay for UBE2L3 activity
7.9 μM ubiquitin,63 nM UBA1 and 25 μM ATP were incubated with different concentrations of UBE2L3 in 10 μl reaction system containing 40 mM Tris-HCl pH 7.4, 20 mM MgCl2, 0.5 mM DTT, 0.1 mg/ml BSA at 37℃ for 1 hour. 10 μl of AMP-Glo Reagent I was added to the reaction and incubated for 1 hour at room temperature. Then 20 μl of AMP-Glo Detection Solution was added and luminescence was read after another 30 min incubation.
Storage
Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
