Immunogen
This PXR antibody was raised against a peptide containing amino acid residues 57-71 of human PXR.
Buffer
Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.
PXR pAb tested by Western blot.
Hep G2 nuclear extract (20 µg per lane) was probed with PXR pAb at a dilution of 1:1,000.
Background
PXR (NR1I2, Pregnane X receptor, nuclear receptor subfamily 1 group I member 2) belongs to a family of transcription factors characterized by a ligand-binding domain and a DNA-binding domain. PXR is an orphan receptor that regulates transcription of the cytochrome P450 gene CYP3A4, binding to the response element of the CYP3A4 promoter as a heterodimer with the 9-cis retinoic acid receptor RXR (RXR-α and RXR-β). PXR is activated by a range of compounds that induce CYP3A4, including dexamethasone and rifampicin.
Positive Control
Active Motif's Hep G2 nuclear extract (Catalog No. 36011) can be used as a positive control for PXR antibody.
