Background

KAT6B (Lysine Acetyltransferase 6B), also called as MOZ2, MORF or MYST4, is a histone acetyltransferase and component of the MOZ/MORF protein complex. It has a histone H3 acetyltransferase activity. In addition to its acetyltransferase activity, the encoded full length protein has transcriptional activation activity in its N-terminal end and transcriptional repression activity in its C-terminal end. So it may be involved in both positive and negative regulation of transcription. KAT6B is necessary for RUNX2-dependent transcriptional activation and could be involved in brain development. Mutations have been found in patients with genitopatellar syndrome. A translocation of KAT6B gene and the CREBBP gene results in acute myeloid leukemias. It may also be involved in cerebral cortex development.

Recombinant KAT6B / MORF (718-1008) protein that includes amino acids 718-1008 of human KAT6B protein (accession number NP_036462.2) was expressed in a baculovirus expression system and contains an N-terminal FLAG tag. The molecular weight of the protein is 35.7 kDa. This product is the catalytic domain of KAT6B protein.

Recombinant KAT6B / MORF (718-1008) protein activity assay
0.5 µg Histone H3.1 (Cat. No. 31296) was incubated with 0 µg (-), 0.1 µg (+), 0.2 µg (++), 0.4 µg (+++) KAT6B / MORF (718-1008) protein in a 20 µl reaction system containing 50 mM Tris-HCl pH 8.6, 2 mM MgCl2, 1 mM TCEP, 0.02% Triton X-100 and 20 µM Acetyl-CoA for 2 hr at RT. Reaction products were detected by Western blot using H3ac (pan-acetyl) antibody (Cat. No. 61637).
