AbFlex® recombinant antibodies are genetically derived from DNA sequences of parental hybridoma clones. For details on the parental clone, see Catalog No. 61273.

Immunogen

This antibody was raised against Bromodeoxyuridine conjugated to BSA (Bovine Serum Albumin).

Buffer

Purified IgG in 140 mM Hepes, pH 7.5, 70 mM NaCl, 32 mM NaOAc, 0.035% sodium azide, 30% glycerol.

 

AbFlex® BrdU antibody (rAb) tested by Immunofluorescence
HeLa cells were stained with 0.5 µg/ml BrdU antibody, followed by anti-mouse IgG2a – Alexa Fluor 488 (lower panel) or DAPI (upper panel). Prior to staining, HeLa cells were treated for 1 hr at 37°C in cell culture medium supplemented with 10 µM BrdU. DNA was denatured wtih NaOH treatment for detection of BrdU incorporation in cells undergoing DNA replication at the time of labeling.

Background

AbFlex® antibodies are recombinant antibodies (rAbs) that have been generated using defined DNA sequences to produce highly specific, reproducible antibodies. Each AbFlex antibody contains a 6xHis Tag, a Biotinylation Tag for enzymatic biotin conjugation using the biotin ligase, BirA, and a sortase recognition motif (LPXTG) to attach a variety of labels directly to the antibody including fluorophores, enzymatic substrates (HRP, AP), peptides, drugs as well as solid supports. AbFlex® BrdU antibody was expressed as full-length IgG with mouse immunoglobulin heavy and light chains (IgG2a isotype) in mammalian 293 cells.

BrdU(Bromodeoxyuridine) is a thymidine analogue, capable of being incorporated into DNA in place of thymidine during DNA synthesis. By pulse labeling with BrdU, the DNA synthesis in the cells can be analyzed by detection with anti-BrdU antibodies. Antibodies against BrdU recognize BrdU in single-stranded DNA (produced by partial denaturation of double-stranded DNA), as well as free BrdU or BrdU coupled to a protein carrier.
