Application Notes

Applications Validated by Active Motif:
WB: 1 µg/ml dilution

AbFlex® recombinant antibodies are genetically derived from DNA sequences of parental hybridoma clones. For details on the parental clone, see Catalog No. 61075.

Immunogen

This antibody was raised against amino acids 1-262 of human c-Myc (myc proto-oncogene protein).

Buffer

Purified IgG in 140 mM Hepes, pH 7.5, 70 mM NaCl, 35 mM NaOAc, 0.035% sodium azide, 30% glycerol.

AbFlex® c-Myc antibody (rAb) tested by Western Blot.
HEK293 cells were transiently transfected with empty vector (Lane 1) or with human c-Myc protein (Lane 2). 10 µg HEK293 whole cell extract was loaded on SDS-PAGE gel and probed with AbFlex c-Myc antibody at 1 µg/ml. MW: 68 kDa.

Background

AbFlex® antibodies are recombinant antibodies (rAbs) that have been generated using defined DNA sequences to produce highly specific, reproducible antibodies. Each AbFlex antibody contains a 6xHis Tag, a Biotinylation Tag for enzymatic biotin conjugation using the biotin ligase, BirA, and a sortase recognition motif (LPXTG) to attach a variety of labels directly to the antibody including fluorophores, enzymatic substrates (HRP, AP), peptides, drugs as well as solid supports. c-Myc is a sequence specific DNA binding transcription factor that regulates gene expression, especially genes involved in cell division. Myc family members contain a basic Helix-Loop-Helix leucine zipper (bHLHZ) domain. Myc family proteins associated with thousands of genes, especially those involved in cell growth and proliferation. Myc serves as an integrator of signal transduction, linking extracellular signals to an acute cellular growth response. Mutations and overexpression of c-Myc are associated with cell proliferation and cancer.
